Reza Yousefi was born in 1972 in Ramian, Gorgan (northern Iran) and completed his education from elementary school to high school in his hometown. In 1996, he graduated in bachelor's degree of biology from Shiraz University, and in the same year, he joined Institute of Biochemistry and Biophysics (IBB) of the University of Tehran to continue his education in master and then in PhD of biochemistry.
After a sabbatical period in Switzerland (Bern University) and passing two postdoctoral periods in Iran (IBB) and France (INRA, Nantes), he was employed by Shiraz University in 2009 and was promoted to professor of biochemistry at this university in 2018. In addition to teaching biochemistry and advanced topics on the structure and function of proteins, his research interests are mainly focused on the application of the recombinant DNA technology for expression and production of the disease-associated or therapeutic proteins and peptides and their structural and functional studies. Also, the production of mutant proteins and peptides with the aim of creating improved products or discovering the relationship between mutations and their pathogenicity is another important research interest which is followed by him. Reza Yousefi is also a member of the editorial board of a number of prestigious international journals.

The Protein Chemistry Laboratory (PCL) at the University of Tehran accepts master's and doctoral students from top universities who demonstrate exceptional talent. Admission is carried out in a professor-centered manner, with the research project emphasizing the expression and laboratory production of therapeutic proteins and peptides in a bacterial host system, along with their structural and functional analyses. Research Interest:
Our research group is interested in the role of mutation and chemical modification on structural/functional variation and fibrillation of eye lens crystallins and human insulin. Crystallin proteins are pathogenic targets for various chemical modifications and mutations which ultimately affect the proper function and transparency of eye lens, resulting in cataract and blindness development. Stability and fibrillation of human insulin is currently a big medical and biotechnological challenge. Site-directed mutagenesis is an important technique to obtain new insulin molecules with increased stability and lower propensity for aggregation. We are also planning to produce novel insulin analogs through bioengineered modifications of the natural amino acid sequence of human insulin. We also use DNA recombinant technology, while using new methods and innovations in optimizing the production of pharmaceutical peptides such as those with incretin activity.

Advanced Protein Biochemistry
The Purpose of the Curriculum
The polymer structure consisting of twenty monomeric building units of proteins (twenty standard alpha-amino acids) and their subsequent folding into a fine three-dimensional structure allows these biomolecules to perform a wide range of biological tasks, from catalytic roles to participation in signaling pathways. In addition to the structural and functional diversity, proteins and their building blocks have been widely used in medical fields and various industries. Understanding this relationship helps biochemistry students to deepen their basic knowledge in the field of protein structure and function, and to learn the important and diverse applications of these biomolecules in different fields. The purpose of this course is to teach graduate students the connection between the principles and scientific rules governing the structure and function of proteins and their practical and applied areas.

(1) Introduction of bio elements in the periodic table, important functional groups, isomerism and non-covalent interactions in biomolecules. (2) Standard alpha-amino acids and biological importance/medical applications of their derivatives. (3) Peptide bond, Ramachandran plot and introduction of medical and industrial applications of peptidase enzymes. (4) Description of the traditional and new methods in determining the primary structure of proteins. (5) Scientific bases of the formation of protein secondary structures and the methods of studying these regular structural elements. (6) Stability of proteins, mechanism and thermodynamics of protein folding. (7) The mechanism of protein aggregation/fibrillation, amyloid diseases and chaperone systems.(8) Different protein families and super families based on biological functions and structural elements. (9) The importance of filamentous and globular proteins in living systems and their medical and industrial applications. Methods in Biochemistry
The Purpose of the Curriculum
The purpose of this course is to transfer the basic knowledge of important and widely applied methods that are often used in biochemistry research to master's and doctoral students of biochemistry.
Syllabus Details
(1) Analytical Methods- Protein, nucleic acid and enzyme analyses. (2) Chromatography Methods - In this section, the principles of size exclusion chromatography (SEC), ion exchange chromatography, hydrophobic interaction chromatography, reverse phase chromatography, affinity chromatography, chromatofocusing, and HPLC/FPLC will be taught. (3) Electrophoresis Methods- In this section, one-dimensional electrophoresis (1-DE) including different types of polyacrylamide gel electrophoresis such as reducing and non-reducing SDS-PAGE⸲ Urea PAGE⸲ native gel electrophoresis, also isoelectrofocusing (IEF) and two-dimensional electrophoresis are taught and discussed along with their important applications. (4) Spectroscopy Methods- In this part, the principles and application of different spectroscopic methods such as UV-Vis absorption spectroscopy, Fluorescence, infrared spectroscopy (IR), Raman spectroscopy, circular dichroism (CD), dynamic light scattering (DLS), mass spectrometry (MS) and nuclear magnetic resonance (NMR) are taught and discussed. (5) Immunochemistry Methods- Principles and important applications of radioimmunoassay (RIA)⸲ western blot and ELISA will be taught. (6) Microscopic Methods- Principles and a number of applications of scanning electron microscope (SEM), transmission electron microscope (TEM), atomic force microscope (AFM) and fluorescence microscope in biochemistry researches are discussed. (7) Thermal Analysis Methods- Principles and applications of differential scanning calorimetry (DSC) and thermal gravimetric analysis (TGA) will be taught. (8) Gene Manipulating Techniques- RNA and DNA extraction, PCR, qPCR and gene cloning.

The Purpose of the Curriculum
Today, protein engineering products are very diverse and they are used in various industries such as pharmaceutical, health, chemical, food, paper, leather and cosmetic industries. The general goal of this course is to acquaint graduate students of biochemistry with the different stages of engineering and production of proteins and peptides. The specific objectives of this course are as following: (1) Teaching common methods and strategies in protein and peptide engineering. (2) Teaching the methods of optimizing the production of recombinant proteins and peptides. (3) Introducing important solutions for the purification of recombinant proteins and peptides and peptides. (4) Introducing pharmaceutical forms and mechanism of action of medicinal proteins and peptides.

Protein Chemistry Laboratory (PCL) is ready to cooperate with various industries by taking advantage of long experience and knowledge in the field of purification⸲ structural and functional studies of proteins and peptides. Our team members possess proficiency in diverse methods of producing and purifying proteins and peptides on a laboratory scale, both from natural sources and using recombinant DNA technology in the bacterial host system. Furthermore, we possess a comprehensive understanding of the structural, stability, and functional assessments of proteins and peptides. We welcome the opportunity to engage in industrial cooperation and look forward to sharing our knowledge and skills to advance the goals of our partners in the industry sector.
